  With the increasing amount of biological data, the need for computational skills has become prominent. Interdisciplinary fields such as computational biology require interdisciplinary collaborations; in order to create an environment for sharing skills and experiences, opportunities for funding and training, career path advice, and research ideas. Creating such environments also help to establish an inclusive platform where students and early-career researchers in computational biology can socialise and help each other in an informal setting. Currently, this is achieved locally at department level at a university or industrial company. The social interactions between students and established scientists are, therefore, consequently limited to the geographical locations of their institutions. The Regional Study Group Norway (RSG-Norway), part of the International Society of Computational Biology (ISCB), aims to create a country-wide platform, to bring together aspiring and established bioinformaticians and to develop an active presence in the Norwegian bioinformatics community. Regional level communities such as RSG-Norway are particularly helpful for students to broaden training and career opportunities. It can also help foreign researchers and students, in settling as new arrivals, through building contacts with those who are familiar with studying and working in Norway. Finally, the study group can be a channel for students and trainees to initiate and/or realise their own ideas regarding social and professional communication with fellow bioinformaticians. RSG-Norway works together with Nordic Computational Biology (NCB), which serves as a resource and knowledge-exchange hub to bring together people from industries and academia as well as local communities such as Regional Student Groups across the Nordics. As part of the NCB, Nordic RSGs are able to build cross-border collaborations and organise open events that reach a wider audience strengthening the community's knowledge-base. The presentation for this abstract will include information on past events and initiatives by RSG-Norway and NCB, their future activities and plans, and how to get involved as a member and an advisor to help develop the computational biology community in Norway and the Nordics.

  The spring wheat cultivar ‘Naxos’ shows high levels of partial resistance to powdery mildew. QTL mapping using SSR markers in the population ‘SHA3/CBRD’ x ‘Naxos’ identified resistance loci from ‘Naxos’ on 1AS, 2BL, 2DL and 7DS (Lu et al 2012, TAG 125: 297-309). The objectives of the present study were to saturate the ‘SHA3/CBRD’ x ‘Naxos’ SSR and DArT marker maps with SNP markers from the Illumina iSelect 90K wheat chip, perform new QTL mapping to confirm previously detected resistance loci contributed by ‘Naxos’, and possibly discover more QTL. A second objective was to validate the powdery mildew resistance QTL from ‘Naxos’ in a new Genetic background and identify tightly linked SNP markers. In the 164 F6 RIL population ‘SHA3/CBRD’ x ‘Naxos’, 9,229 polymorphic SNP markers were added to the previous linkage maps. QTL mapping with the combined maps identified previously detected QTL from ‘Naxos’ on 1AS, 2BL and 2DL. In addition, a QTL contributed by ‘Naxos’ was detected on 2AL and a second one detected on 1A. For confirmation of the detected QTL, the 131 F6 RIL population ‘SORU#1’ x ‘Naxos’ was evaluated and 10,372 polymorphic SNP markers from the 90K wheat chip were used to develop linkage maps. QTL mapping in this population confirmed the resistance QTL on 1AS and 2AL contributed by ‘Naxos’. The SNP markers identified in this study will be useful for breeding cultivars with improved resistance to powdery mildew.

  Pre-harvest sprouting (PHS) is the most important quality-degrading factor of wheat in Norway. PHS is caused by the untimely germination of grains before harvest, while they are still on their mother plants in the field. This leads to production of alpha-amylases, proteases and glucanases that break down cell walls, starch and gluten proteins in the endosperm, and destroys the breadmaking quality. PHS occurs in susceptible cultivars when exposed to rain after maturity and is to a large extent determined by the level of seed dormancy, a complex quantitatively inherited trait that is highly dependent on environmental conditions, genetic background and genotype-byenvironment interactions. However, despite the high importance of this trait, little has been known about the genetic basis for PHS resistance in the Norwegian wheat germplasm until recently. Wheat genomics resources, including high-density SNP chips and genome sequence data has facilitated big advances in the molecular genetic understanding of this trait. This talk will give a review of recently cloned genes for seed dormancy in wheat, and how this relates to PHS resistance in Norwegian germplasm. The recently funded project “Expanding the technology base for Norwegian wheat breeding: genomic tools for breeding of high quality bread wheat” (NFR 256370, 2016-2020) will also presented. In this project, Graminor, NMBU and NOFIMA are joining forces to unravel the genetic basis of PHS resistance in Norwegian wheat, develop molecular markers for the trait, and lay the grounds for applying genomic selection in Norwegian Wheat breeding.

  BAC libraries for chromosome 7B were constructed with an insert size of 185 and 120kb for 7BS and 7BL respectively. Fingerprinting has been completed for both arms. For 7BS the a minimum-tiling path (MTP) consists of around 4000 BAC clones, while the 7BL MTP consists of around 6000 BACs. Sequencing of the individual BACs of both arms are under way. A chromosome 7B radiation-hybrid panel is currently being characterized using ISBP and other markers markers derived from the 7B survey sequencing project. In parallel to our genomic sequencing and mapping efforts, we are building a cell specific, homeologue specific transcriptome map of CS spring endosperm at different developmental stages.

  Construction of physical chromosome maps is dependent on accurate ordering of BAC contigs using genetic maps. Such genetic maps require very high resolution along the entire chromosome, which is difficult to obtain by conventional recombination-based genetic maps. One of the alternatives is to generate high-resolution genetic map with the use of a radiation-hybrid (RH) panel. The focus of this study was to develop a CGH NimbleGen high-density microarray chip for genotyping RH-population and construction of a high density RH-map of wheat chromosome 7B. Seven hundred and fifty thousand probes of 50bp were designed from 7B shotgun assemblies and printed on a 3x720K NimbleGen chip. These probes represented a mixture of ISBP markers and random genomic fragments (RGF). Genotyping of the parental lines of the RH population, i.e. the durum wheat Langdon (7B+) and the Langdon 7B-7D substitution line (7B-), identified 44.035 RGFs and 5.465 ISBPs showing presence-absence variation between 7B+ and 7B- lines. A re-designed 12*270K chip containing four replicates of the 49500 probes was subsequently used to genotype a subset of wheat 7B deletion stocks and allowed assignment of markers to the deletion bins.

  The development of molecular markers in wheat has always been a formidable task due to the massive, complex and polyploid genome. The hexaploid wheat genome is about 17Gb. Significant progress is now being achieved in assembling the DNA sequences from individual chromosome arms of Chinese Spring. These assemblies provide an opportunity to enhance the analysis of wheat genome and surve a template for developing the functional markers in wheat. The results reported here are part of a larger project whose main goal is the construction of an integrated genetic and physical map of wheat chromosome 7B.   The international wheat genome sequencing consortium (IWGSC) aims to provide a publicly available reference sequence of the 17Gbp hexaploid wheat genome through sequencing of single isolated chromosome arms. Norway is contributing to this work by sequencing chromosome 7B. Here we outline our work towards a 7B reference sequence and beyond. Sequencing and assembly of 7B has been carried out using two different strategies; whole chromosome arms shotgun sequencing and BAC-by-BAC shotgun sequencing of ~10,000 BAC clones representing the minimum tiling path (MTP) of 7B. BAC-by-BAC assemblies demonstrated a great improvement of assembly quality compared to shotgun assemblies of whole chromosome arms. The MTP of 7B consist of ~500 BAC-contigs, and to order these along the chromosome, we first used CGH-based genotyping of presence-absence variation to anchor ~4000 sequences to 9 chromosome deletion bins. Further fine-ordering of BAC-contigs within deletion bins is ongoing using genetic maps based on a radiation-hybrid panel. In parallel to our genomic sequencing and mapping efforts, we are building a cell specific, homeologue specific transcriptome map of endosperm at different developmental stages by integrating the ongoing genome sequencing resources.
